MECHANISM OF ACTION 

The core antifungal pathway is as follows:

  • Itraconazole targets fungal cytochrome P450-dependent enzyme lanosterol 14-alpha-demethylase. 
  • It stops the chemical step that is needed to make Ergosterol. 
  • Since there is no conversion of Lanosterol to Ergosterol, it causes holes and leaks in the fungal cell wall membrane. 
  • Lastly, there is fungal cell death.
